By Bus From Manila:  

Buendia or Cubao terminal, board a bus bound for Lucena. DLTB and Jam Liner. These buses usually pass by Santo Tomas, but to be safe, ask the conductor to drop off at Brgy San Pedro going to Padre Pio Shrine. 

at San Pedro where ride a tricycle to Padre Pio National Shrine of St. Padre Pio (or just say Padre Pio). You can hire the tricycle for a special trip.
